What about Johto? and Pokemon Contests? Is this game a straight update of the first two?  If so what about the Pokemon Nintendo locked away from us when GSC were made incapatible with Ruby and Sapphire?  Do they appear in Kanto now?

The GB/GBC Pokemon games aren't compatible with Ruby/Sapphire because it's not technically possible to link up Game Boy to Game Boy Advance.  They use different data transfer protocols.  As for why the original monsters weren't just included in R/S, my guess would be a combination of memory limitations and because they were already planning Fire Red and Leaf Green.  You should now be able to trade Pokemon among all of the GBA games, as well as Pokemon Colosseum, although Ben would have to confirm that for me.

If you watch the trailer for Colosseum, you'll notice it has many of the GSC pokemon in it.  I'm guessing it was planned on nintendos part to have a combination of RS, RG, and Colosseum allow you to catch all of the pokemon.

Whoa, whoa, slow down!  First off, Johto is not accessible in Fire/Leaf, although:

There are extra islands to travel to after beating the Elite Four.  They weren't in the original and they contain SOME (not ALL) Pokémon from G/S/C, along with a breeding center.  Once you complete RPG Mode in Colosseum, you can trade over the Pokémon you've captured from there to your GBA games.  There's more stuff on the Fire/Leaf Islands that I won't reveal here.

So, to answer your question, no, the game is not a straight update of the first two.  It's more like an 'upgrade'.  The original game is there, but there is new stuff too to keep owners of the original games on their toes.

There are no Pokémon Contests in Kanto (I could be mistaken, but I'm fairly sure about this), but I don't see why you can't enter your Kanto Pokémon in Hoenn Contests by transferring them over.  I doubt it would be a problem.

Yes, aside from graphical enhancements and some minor tweaks such as more trainers along routes, some items replaced with others (ie. there's no more 'HP up' in Mt. Moon anymore), two on two battles, the levels are still the same.  Not sure if any two on two battles take place in the gyms yet.  Because I'm crap.

EDIT: Sup Ben we have "spoiler" tags.

Thanks Aussie Ben.  You probably haven't had the opportunity to do much trading, but I know when I used to trade alot I got angry when my pokemon didn't remember who its original trainer was and forgot who was feeding it (You know tameness).  When you get the chance could you tell me if Pokemon retain their memory of their old trainers or better yet if their tameness stays the same when you trade them.

I think it would be cool if nintendo allowed for straight transfers, or do input your same name and id number in a new game so you would still be the "same" trainer.

What about the moves? Are there moves from S/G and R/S or is it the same moves from the original R/B?

Since they connect with Ruby and Sapphire, I'm asuming they updated the move list, but that brings up an interesting point.  Did GameFreak add any moves or new mechanics that were previously unseen in Pokegames.
Oh is the TM list the same as the first Red and Blue/Green?
