The third Japan-only Taiko Drum Master game for the Wii will include a track from New Super Mario Bros. Wii, among other video game songs. When playing the game, the background will feature Mario and Luigi dancing along with other characters such as the Koopa Kids.

Other video game cameos in the game include the Idols from Namco's Idol Masters series and characters from Capcom's Monster Hunter series. Video game tracks include God Eater, Ridge Racer Type 4, and Nintendo's Pokemon Diamond and Pearl. The final track list will include 70 songs.

In addition to the songs, seven new mini-games were revealed. These mini-games use the rhythm gameplay mechanics to achieve goals. The first of these is Wani Wani Panic, where players bang on the drums in order to scare away alligators. In the second game, Haunted House, players guide the characters through a haunted house and must perform well in order to protect them. In Pirate Don-Chan and the Secret Treasure, players take on a giant squid and collect the treasure.

Alien Attack has players co-operating to bang the drums in unison in order to unleash a powerful beam. To The Heavens: Cake Tower is a two-player co-op game where players climb a cake to the top. Great Honeybee Operation has players controlling bees. The first player to tap a flower will receive the honey.  The final game is Fairyland Pumpkin Derby, this is a racing game where players control a pumpkin carriage.
