The karaoke revolution games that have two-player modes always have a small handful of songs labeled as "true duets," which for some songs was a simple male-lead and female-lead division, but for some more interesting songs, had you singing lead and backup parts, like in "Uptown Girl," "Dust in the Wind," and "Twist and Shout." But yeah, each player was scored based on their pitch, there was no kind of bonus for how your voice related to the other person's voice, I guess.
I do agree that this would be a good game to implement a second microphone, and/or a third guitar.
One other thing, that "45 songs" number was dismissed by Harmonix as speculation.
