I wonder how long it'll be before a 3rd party creates a little red laser sight for the Wii Controller?

For third person shooters, you'd just need a virtual laser, like Resident Evil 4.  The Wiimote will never be perfectly calibrated to the screen in a way that a laser dot will line up to the in game target.  I suppose a light gun style game could be made, but that would require a special sight adapter on the controller anyway.

It would be easier to just have the game put a red dot on the screen where the laser is pointing.

Lasers are hardly visible when striking a screen, because glass doesn't scatter the laser.   And the laser would never be in the right place anyway.  I don't thing you understand how the Revolution works.  The controller isn't a light gun, and it doesn't emulate a light gun.  (I suppose it might be able to with a scope add-on and some calibration)

I'm assuming that somehow, you'll have to calibrate the Wii Controllers anyway, right?

I think calibration will varry depending on the game.  Some won't really need it.  Some will just detect where the sensor bar is.  With some games you'll have to hold the controller in a comfortable nuetral position and press the home button. I don't think it will get much more complicated than that.

Glass is reflective. There's a warning on pretty much every laser pointer "don't point at reflective surfaces".
